Getting prayer, getting there, getting back:
I use digsite necklaces to get an extra prayer boost from the Temple of Zaros. After looting the chest you can tele back to the digsite for a recharge. Draken's medallion is good if you have it (make sure to keep it charged). Using this strategy I often complete the barrows without using a dose of prayer or using only 1 dose, and can usually make 2 or 3 runs before banking.
Alternatively one can use a ring of dueling tele to Mobilizing armies , run to Oo'glog and use the sulphur pool for a prayer boost, take a relaxing salt water bath, and use the bank. If you don't have Draken's you can then tele to house and thence to Canfis to get back.
I always start with Dharok then fight 1 or 2 others, using magic, till my prayer is about half gone, before taking Ahrim with a crossbow. After Ahrim use a super restore which will give a prayer boost as well.
I use full Ahrim's set plus Karils crossbow to fight Ahrim. I then switch to full Guthan's when I enter the tunnels.
Revision:
From reading these posts I see that using mage armor is unnecessary so now I just use Guthan's with Karil's Crossbow and a Mystic fire staff to cast fire surge, using Guthan's Warspear in the tunnels. This gives me 3 extra inventory spaces and allows more Barrows runs between banking. I have just competed 5 runs before returning to the bank for another digsite necklace, and I could have done more.

Here's a tip I don't think has even been mentioned yet: use SafeSpots if you are using magic and range against the meleeing brothers!
Verac, Dharok, Torag, Guthan, and Akrisae can all be safespotted so you don't have to use any food/potions except for the 1-2 hits they might get when you lure them
Each of their rooms has little rubbles of rock jutting out.. you just open the tomb from the opposite side and get each of them hooked on the rubble and voila, you are safe from attacks!

every brother has a weakness according to the combat triangle, so many players bring a spell to cast against the four melee brothers (slayer dart or the polypore staff's spell are the most efficient) and then use melee against ahrim and karil.
the single most valuable quest item to have for this minigame is drakan's medallion from branches of darkmeyer. It allows you to teleport straight to the barrows, among other useful places (like the burgh de rott bank!)
Using drakan's medallion in combination with the ardougne cloak's altar teleport is a quick way to recharge prayer inbetween runs. Simply teleport out with the cape upon looting the barrows chest, and teleport right back with the medallion. Don't forget to recharge your medallion if you use this method
After you complete Ritual of the Majarrat, you unlock a seventh brother, "Akrisae the Doomed". Killing all 6 + akrisae does NOT increase your chance of getting a piece of barrows armor further, so instead of killing all 7 each run, you can skip any one of them and still have the maximum chance of a barrows armor piece
You don't need a spade anymore! The creepy old tender of the barrows has them set up for you.
Akrisae switches protection prayers after taking a certain amount of damage (200 or so) from one style. Have a setup that allows you to switch attack styles quickly to kill him. I use full void and simply switch out my hat and weapon, however a salamander also allows access to all 3 combat styles easily.
Headcanon Haven, where everything is made up and the points don't matter.

If you don't have Draken's amulet from the Branches of Darkmeyer, Get the character Zachory Bragg to level 99 in temple trekking and toggle the option to use the shortcut behind the tavern in Canifis to go straight to the swamp boat. If using house teletabs and a portal in your house Kharyll, Barrowing and Banking can be done if 30 seconds!

Using salamanders is an excellent way to quickly defeat the melee based brothers (they are weak to magic), as it ignores your magic attack bonus. This allows you to wear strong melee armor without worrying about high negative magic attack, so you can defeat all the melee brothers (except Dharok) without using prayer potions. You should have at least 70 defence and monkfish or better food to do this. Just remember to watch your Hp!

In my opinion, the best way to beat them while also spending the least amount on supplies is as follows:
High Levels:
GEAR
Verac's Full Helm (defense and prayer)
Bandos Chest (defense and strength)
Bandos Tassets (defense and strength)
Steadfast Boots>Dragon Boots (Strength)
Goliath>Barrows Gloves
Ring of Vigour (strength and special)
Amulet of Fury
Firecape<>Soul Wars Cape (matter of preference of strength or prayer)
Chaotic Maul>Saradomin Godsword>any other weapon with high crush (crush has an advantage against plate-style armor, the strength bonus of the above listed weapons also have a high enough strength bonus to easily dispatch Ahrim and Karil)
SUPPLIES
One of each: Extreme Attack, Extreme Strength, Extreme Defense (overloads are probably not the best idea because of the length of time sometimes needed to get between crypts, and also through the final catacomb)
Two Super Prayer Restore potions
Three to five rocktail/sharks
(I am including Herblore when I listed this as "high levels" as you'll need at least 94 Herblore to make the Super Prayer Restores)
For players who have the money to spend on the gear listed above, they will; in the end, save quite a bit of money on other supplies, such as runes (for Slayer Dart method) and will create a higher profit margin in the end.
Lower Levels (or welfare)
GEAR
Verac's Full Helm (defense and prayer)
Verac's Brassard (defense and prayer)
Verac's Plateskirt (defense and prayer)
Dragon Boots (strength)
Barrows Gloves (strength)

Verac's Flail OR Bandos Godsword OR a combination of the two (the completion of Verac's set allows for the special which has the chance to hit through armor, PLUS it has a crush attack which is effective against plate-style armor)
