Quick prayer saving tip.
Dont pray when fighting ahrims, Most of his attacks are weakening spells so use your specials against him. You'll be surprised how little he use's aggressive attacks.
Only use prayer for dharoks.
Karils can be deadly, but using good range defensive armour reduces the hits.
I only use 1 dose of pp per run.

Read through quite a few tips from others and couldn't help but laugh at some "you must use prayer" lol. So thought I'd put two.
Tip 1:
Ensure you make profit every trip without counting barrow drop - this should be an extra. (So plan your resources to cost under 50k per chest)
This can be done through using orange/red/black salamander and cheap armour keeping a mage bonus higher than -20, or Using full veracs for higher players.
Tip 2:
Know your enemy - Each brother has a max hit, there are puzzles on the doors, you may not need pray depending on your defence (70 is sufficient to only pray dharok and karil) - therefore use starting pray on these two first- carrying 1 pray pot incase one is within the tunnel or you run out of pray. Extra resources should be carried upon early trips to get the hang of it. Reading up on barrows before hand is vital - otherwise you will waste time and money.

Utilize the combat triangle! I've found that using slayer dart (or for the higher levels, a polypore staff) is perfect for the melee brothers, and bring a melee weapon for Ahrim and Karil.
Remember that you loot mind and death runes from the chest, so ideally, you will never run out of slayer dart runes.
Wear prayer-conserving armor, even if you are maging. Keep your magic bonus at -20 or more, and you will still decimate the brothers. Yes, that's negative 20.
For sake of making several runs per trip, bringing a set of guthan's armor is choice. This is because it eliminates such a need for food. You can always heal in the tunnels. (Of course, bring some rocktails for emergency.)

For lower levels (and some higher levels), knowing the right path is crucial, and sometimes, finding that right path is the hardest part. But not only is the path important, but the inventory setup is also very variable depending on the person.
I can't give any specifics to inventory setup, since one person might be a little bit slower than another, but I can give basic necessities.
Prayer potions (2-4, 4 doses each)
Super set/Extreme Set/Overload (1 set)
Two different styles of attack (Mage and Melee recommended)
Runes (For teleports, and freezing [if on Ancient's, or using Snare on Regular Spellbook])
Backup armour and weapons (Having two types of armour specializing in defence and attack respectively helps when killing, and travelling the tunnel)
Food (Required, if planning to do multiple loots in a single trip, or you run out of prayer, and don't wish to use more prayer potion doses)
Path (This is based off of my own experience, and what I find to be the optimum path):
1) Karil- Karil uses Ranged, and can hit very hard, if you don't have protection prayers on. He attacks rapidly and accurately with his repeater crossbow, and can hit in the 200s on even the best of people. Prayer for him is almost necessary. Many people choose to use melee on Karil.
2) Dharok- Since Dharok is the highest leveled brother, and can also hit the highest with his Wretched Strength ability, having Protect from Melee/Deflect Melee is a necessity. Some people choose to freeze him/bind him using Snare, or Ice Burst/Blitz/Barrage. After using that, they continue to attack him with magic of their choice, such as Polypore Staff, more spellbook spells, or a Salamander.
3) Verac- It may seem like using Prayer on Verac may be a waste, but it does help. His ability only hits through your prayer on occasion, meaning the rest of the time, his attacks are blocked. Like Dharok, and the other melee brothers, Verac is weak to magic, so using a method similar to killing Dharok is used often.

4) For those who have beaten Ritual of the Mahjarrat, you have unlocked the 7th Barrows Brother, Akrisae. Now, contrary to his armour set consisting of robes, Akrisae uses Melee when fighting. So using Protect from Melee/Deflect Melee will fully negate his attacks. However, killing Akrisae is different from the other Brothers. Akrisae uses Protection Prayers that correspond to the attack style you are fighting him with. So two attack styles are a must when fighting him. A common method for people is to use a Korasi Sword on him, use melee until he switches to Protect from Melee, use the Special Attack, which hits with Magic, so that Akrisae with switch to Protect from Magic, continue attacking with melee, and if you have Recover Special Potions, you may use that. Other than that, many players just use melee and Ranged to kill him. Akrisae has the ability to occasionally deactivate your Prayers, and even drain them, should he be allowed to hit you. He has a max hit of 340, so watch yourself!
5) Torag- At this point in the trip, you only have 3 brothers left, and two of them are melee brothers, which can all be killed similarly. Players just tend to kill the closest brother to them at the time, in this case, Torag. He uses Melee, and can drain your energy (Quite a useless ability, I know). Like Verac and Dharok, he is weak to magic, but out of all the brothers, Torag has the highest Melee defence, so using melee on him is not recommended, unless you are using a Verac's set. Torag has the ability to hit 200+ on occasion, so if you do get hit for this, do not freak out.
6) Ahrim/Guthan- It doesn't matter which one you kill at this point, it'll all have to do with preference. Ahrim used Magic, and can easily be blocked without prayers using Void/Dragonhide. Ahrim can lower your melee stats, you can choose to try and kill him quickly with melee, or use Ranged. Guthan uses melee, and is also weak to magic. He can heal the damage he hits you for on occasion.

Learn how to use prayer!
There is absolutely nothing worse than getting Dharok down to a sliver of health and his set effect kicks in and starts taking chunks off of your health like it was nothing. Of course leave Verac as the last brother because he hits straight through prayer, so if you are using prayers they have more than likely been drained at this point.
Know the Answers
In the Barrows Crypt if you get the answers wrong it resets everything and you have to figure out where you are going to go and then answer another question, get it right the first time or your health and prayer is going to be wasted in the tunnels.

My own personal setup and tips.
I've been doing Barrows for a good number of years, so I've learned a lot of tricks to killing the brothers. Remember, this is my own setup, yours does not have to be exactly like this.
Setup (Inv and equipped armour):
Ahrim's Top and Legs
Polypore Staff
Dragon Dagger p++
Crystal Bow
Vecna Skull (I only have 92 Magic, so I boost to 94+ for Ice Barrage)
2k Water, 1k Death, 1k Bloods, and 20 Law runes
1 Recover Special Potion
1 Super Set (No Defence Potion)
2 Prayer Potions
12 Sharks
Armour:
Neitiznot Helmet
Amulet of Fury
Rune Platebody and Legs
Dragon Boots
Chaotic Longsword
Dragon Defender
Soul Wars Cape
Ring of Wealth
Goliath Gloves
Reverence

Personal Path:
1) Karil
2) Dharok
3) Ahrim
4) Verac
5) Torag
6) Guthan
After killing all the brothers, I either choose to backtrack and do another loot, OR, I teleport to Lassar(On the Ancient's Spellbook) which takes me to Ice Mountain, I run to the Monastery to get healed by the monks, and restore my Prayer. After that, I teleport to Kharyrll (Canifis), to bank, restock my inventory, and head out for another trip.
If you have Drakken's Medallion, you can just teleport to Darkmeyer to restore your Prayer, then teleport to Burgh de Rott to bank, then teleport straight back to Barrows.
Tips:
For those using Ancients, and have access to Ice Burst+, using those with a mix of another distance method on the melee brothers very much helps. When using Ice Burst, you are able to walk away from them, and begin attacking them from a distance for a short amount of time.
Ice Burst- 3-4 Hits before they begin moving
Ice Blitz- 5 hits before they begin moving
Ice Barrage- 5-6 hits before they begin moving
When in the Chest room, there are two candlesticks to the south of the Chest. IF you have a melee brother in the room, you can be able to freeze them and begin attacking them from the other side of the candle. Just use the number of hits you can before they'll start moving, wait, then repeat.
