Well, in terms of general drawbacks that a sibling might disapprove of, everyone suffers a gradual attitude change. The sibling will catch on to this, and start to ask questions like: "Why you being a dick? Why did you attack that guy when you could have asked him to move out of the way? You're disappearing for long hours and when I asked you what was going on you growled at me. Also I don't like that you are basically a slave to some shady guy who can kill you whenever he wants." And then come to the conclusion that the powers are bad.
Also I'd say that the Vampires would be better off in the sense that they attract the least attention. They favor stealth and manipulation through their Invisibility and Charm, none of which actually require them to transform. If they do get detected, they can use their invisibility to escape, transform and fly away, or talk their way out.
Meanwhile the Champions are walking beacons of light, and the Werewolves are loud and destructive. Their abilities are more combat oriented, and neither of them are very subtle.
Also remember that your goal isn't just to wipe the others out, but to make your own presence stronger. Vampires can charm their way through the law enforcement, the city's government, and the upper class, which is great for their ultimate goal of taking over the city.
TL;DR: While the Werewolves and Champions are busy fighting in the streets, the Vampires gather recruits and slowly turn the city's elite. The numerical advantage combined with their ability to stay hidden and the influence at their disposal will make them a force to be reckoned with, if they play their cards right.
