back in the day, 2006ish, when everyone and their dog had an account, most of those accounts were ftp, making the ftp characters very memorable
Since everyone went though the tutorial and vannaka is the only guy in it who really makes an impression on you, that makes him quite iconic

I would say duke horacio, elvarg, king roald, count draynor and delrith are also quite iconic. You might never have heard of zanik or azzanadra (even though they're a major part of the game) but you'll have heard of these chaps

also those goblin chiefs... bentnoze and wartface... and obviously the 3 main gods

Sliske is obviously one of them in the modern days.
Hazelmere was pretty big, and Jagex gave him a... sad finale, I guess?
Nomad is obviously big, they made a cosmetic override of his gear and walk animation.
I'd say V was a big mystery guy after WGS came out, A LOT of people were talking about him.
Drakan was another one of those big mysterious figures for the longest time, too.

Depends on what you mean by "iconic". When I hear "iconic", I think of characters that nearly everyone--even people who aren't fans--would recognize. For example, Mario and Pikachu are iconic even though, in the bigger picture of Pokemon, Pikachu isn't all that important.

Characters I consider iconic:

WOM
Saradomin
Zamorak
Guthix
Lucien
Hazeel
Elvarg
Azzandra
Sir Tiffy

Characters like Sliske, Zaros, Seren, and Jas are very important, but I honestly wouldn't consider them iconic. The only one that even has a chance of being recognized outside of RS is Zaros.
Runescape doesn't need a hero...it needs a villain. An all encompassing force of evil that will remain ever-threatening and use chaos to make the peoples of Gielinor tolerate each other, grow strong together, and fight side by side against this evil. I am that villain.

Hazelmere is obviously my personal favorite as I've named my character after him. He's simply the best. The bluntness, wisdom, meditative trance he'd ascend to, even being a skilled mage and huge inspiration for the character.
I'd also say Zanik, Arianwyn, Kuradel, Juna, Nomad. All of the GWD bosses have become quite prominent, and the gods, if you can count them. I enjoy a lot of them.
