Do we actually know for sure that Seren and Guthix were the only ones here in the First Age, or is that purely an assumption? It makes sense if Guthix going to sleep was in fact what marked the start of the Second Age, but do we know even that?

I believe it was stated somewhere in an older piece of content. I would have to find it though.
However, Guthix and Seren being the only ones makes the most sense, as Guthix did not want gods on Gielinor, and Seren was the only one he had to convince.

In Guthix's butterfly memories, I think he said he and Seren were the only gods - indirectly, when he asked her to join him on sleep to leave the mortals alone to their independence.
But still, there could have been one or few gods able to hide from his sight even then - but I guess they'd have to be very weak for that, to not be sensed by him.

The Origins of Gielinor video seems to back up the idea of only Guthix and Seren being present, the book of the gods doesn't refute this either. Like you said though there could have been minor gods like Karamjan etc

what marked the start of the Second Age, then obviously there would be time remaining in the First Age for gods to come while he was sleeping.
As for the Shaikahan, I'd have to check Tai Bwo Wannai Trio dialogue, but its examine message certainly seems to imply it isn't a lion.

I always thought it was the arrival of Zaros that began the second age, as the second age is also known as the Golden Age of Zaros.

Right they say its lion-like but the lions themselves are odd I mean I remember the first time I fought growler and was kind of amazed that he was the magic user kind of odd, and the Shaikahan uses melee, where prideleaders in the BoL also used Ranged/Melee. So I don't know if it is meant to be an actual lion.
