How does the existence of an afterlife for mortals fit into the bigger picture of the elder gods and the great revision? It certainly doesn't seem like a world the elder gods would have made.
Rumour has it there could be a spirit Elder. Though it could be some form of self organisation for sentience based anima lifeforms or what I like to refer to as complex anima. The fact is that the real whys and wherefores of the afterlife have yet to be answered. Even by6 NPCs I'd say most of it is guesswork.
The purpose of adventure is to shine light into dark places,
Poke monsters with a sharp stick, Then steal anything that isn't nailed down!

Elders weren't expecting sentient life, they blocked it's evolution before and I think they don't know and/or don't care about it now.
So I suppose afterlife and soul isn't Elder's agenda now.
Another question is, what would they do if they care. Eat sentient souls along with their usual anima lunch? Or don't care anyway?
6th is mystery. All 5 Elders are connected to Anima, but they represent 5 forms. 6th doesn't. She can represent something even more vague than 5 universal principles of Elders, like pure existence itself or so. Crazy stuff
Yet I think that humans will play important part, perhaps because their ability to have afterlife. Rocks and plants, usual Anima generators, don't have it. Maybe 6th one is organic sentience and afterlife itself, so she doesn't need to care about planet's anima she needs us
Could her plan conflict with other elders, since they want to destroy planet for anima, which would harm flow of souls to afterlife, agenda of 6th one? Does she want to eat/abuse us, or could it be more like cooperation - she would nurture and heal our souls, if we let our souls to join her and help her to exist through us?

It isn't.
We made it ourselves.
Nomad's Elegy seems to lay it out pretty clearly: The afterlife is a world
literally built OUT OF wishful thinking.
We want there to be an afterlife, so there's an afterlife that reflects what we want, powered into existence by the anima in the souls of mortals. The only restriction is that you need to have a soul that passes on in the first place to participate. (So the mahjarrat, who are still in their gems, and gods whose souls turn to pure elder energy relinquish that)
My headcanon about the afterlife is that when a soul dies they actually rejoin the Anima Mundi and the afterlife is some metaphysical construct dreamed into being by all the souls there.

I have noticed your kind does tend to blindly stumble forward towards danger simply because it exists. What is your word for that?
