This is what is often referred to as Gameplay and Story Segregation. The reason King Roald, Captain Rovin, and other important people in Varrock keep asking you to help them with Varrock's problems despite your killing Varrock guards is because your killing those guards isn't part of the story - it's just a gameplay mechanic. They're attackable because they have an 'Attack' option, and you get XP for it just as would anything else you are allowed to attack.
You're also not required to attack the guards at all. You can get XP from giant rats, bears, and various undead creatures in the Varrock sewers if you prefer. So while there might be occasional jokes about how guards have a short life expectancy, no NPC will call you out on killing them yourself within the story, unless there's some 'dialogue choice' option where you either confirm or deny that you have done stuff like that.
But anyway, it's not a canonical part of the story. So outside of a few jokes, it's not even mentioned in NPC dialogue, and certainly does not play any part in whether or not they want to work with you.
