An invasion now would make little sense.
It would make so much more sense for Misthalin to seek rapprochement with the Vampyres, especially now that human blood is less relied on.
It will take time, but if Vanescula is as sincere as she says about honouring the current alliance, now would be the perfect opportunity to start laying the foundations for a total ceasing of hostility, although as I said, it'll take a long time and patience.
The religious aspect is mostly redundant now, Vampyres mostly don't care about Zamorak so much anymore, so as long as the Saradominists are able to restrain themselves from proselyting, I feel they can start by establishing a trade treaty between Varrock, Burgh de rot and Darkmeyer.
Misthalin can export valuable supplies such as metals, fabrics, crops (for the human residents), timber and various other materials that would otherwise be difficult to obtain in The swamp lands.
These can be used to help rebuild Burgh de rot, becoming a trade port for the humans between Misthalin and Darkmeyer, as well as to help gradually improve lives in the vampyre slums, where human ex-cattle can stop being drank from, and start building up into a Civilised town again.
Luxury goods can also help to perhaps further give incentive to the Vampyres to stop and reduce drinking humsn blood.
In exchange, the Vampyres can start trading rare ores only found there, trade blood runes providing a massive boon to human magic institutions and also offers up a new sea-trade route to both Misthalin and the Eastern lands, which potentially means the Vampyres could build themselves a big economic hub if they play ball.
It will take work, but a peaceful resolution is vastly more profitable and safer, not to mention it doesn't waste manpower that could be used in the efforts against the Elder Gods.
